Assessment of genetic diversity among Pakistani wheat (Triticum aestivum L.) advanced breeding lines using RAPD and SDS-PAGE Electron. J. Biotechnol.

Genetic diversity was assessed among 32 advanced wheat breeding lines included in the National Uniform Wheat Yield Trials (2006-07) of Pakistan using molecular (DNA) and biochemical (SDS-PAGE) markers. Of the 72 RAPD primers used for initial screening, 15 were found polymorphic. A total of 140 bands (61.4% polymorphic) were generated by the 15 random decamer primers. Genetic similarity coefficients ranged from 0.81 to 0.94 for rainfed and from 0.70 to 0.93 for the normal seeding date group. Cluster analysis using the unweighted pair group method of arithmetic averages (UPGMA) clustered the 32 advanced wheat breeding lines into one major and three small groups. Caracterización de las inmunoglobulinas del ñandú (Rhea americana) InVet

El objetivo de este trabajo fue caracterizar las inmunoglobulinas séricas y la IgY de la yema de huevo de ñandú, a través de las técnicas de electroforesis en acetato de celulosa, electroforesis en gel de poliacrilamida y Western-blot. Se estudiaron 19 ñandúes clínicamente sanos de 14 a 15 meses de edad, provenientes un criadero de Balcarce, Argentina. La electroforesis en acetato de celulosa de los sueros mostró la presencia de cuatro fracciones proteicas: albúmina, alfa, beta y gamma globulinas. La concentración de las gamma globulinas fue de 0,69 ± 0,18 g/dl, con un rango de 0,42-1,05 g/dl. La corrida electroforética de la IgY muestró que esta se ubica en la fracción gammaglobulínica. Characterization of cry2-type genes of Bacillus thuringiensis strains from soil-isolated of Sichuan basin, China BJM

Sichuan basin, situated in the west of China, is the fourth biggest basin in China. In order to describe a systematic study of the cry2-type genes resources from Bacillus thuringiensis strains of Sichuan basin, a total of 791 Bacillus thuringiensis strains have been screened from 2650 soil samples in different ecological regions. The method of PCR-restriction fragment length polymorphism (PCR-RFLP) was used to identify the type of cry2 genes. The results showed that 322 Bacillus thuringiensis strains harbored cry2-type genes and four different RFLP patterns were found. Characterization of onion (Allium cepa L.) varieties based on physical properties and seed performance Agrociencia

Abstract: Five cultivars (Cristal White Wax, Toro, Red Burgundy, Cojumatlán and Contessa) of onion (Allium cepa L.) were characterized according to: the weight of 100 seeds, germination kinetics, speed germination (T50) and seed protein analysis. Seed of Crystal White Wax was differentiated from the other cultivars by its low weight (0.30 g), since the weight of the rest was 0.40 g. Crystal White Wax had low seed germination (56%) and the highest delay of germination (T50 values were 78 h), while T50 of the rest was 52 h. Total proteins and esterases from silk gland extracts of Bombyx mori silkworm were characterized and compared with electrophoretic profiles of prepared extracts with silkworm glands infected with nucleopolyhedrovirus (BmNPV). SDS-PAGE (7%) gels was used for the total proteins, and 10% native PAGE for esterases. In the silk glands extracts of healthy silkworms, it was observed seven protein zones with molecular weight varying between 10 kDa (P1) and 60 kDa (P7). Comparison of electrophoretic protein profiles of Campylobacter jejuni subsp. jejuni isolated from different animal species BJM

Electrophoretic protein profiles of Campylobacter jejuni subsp. jejuni strains isolated from feces of seven animal species, including man, were compared. Fourteen strains (two from each species) plus two human strains and the reference one, were ruptured by ultrasound and their total soluble proteins were analyzed by SDS-PAGE technique in a 12% polyacrylamide gel with computerized densitometric reading by the molecular analyst software. Comparison of protein profile of co-existing Fasciola hepatica and Fasciola gigantica parasite in Bos taurus (cattle) and Bubalus bubalis (Philippine water buffalo) OAK

Fasciola hepatica and Fasciola gigantica habitually co-exist as parasites of cattle (Bos taurus) and water buffalo (Bubalus bubalis), and despite variation in their morphometry, their species status is suspicious. Liver flukes isolated from cattle and water buffalo were initially sorted as F. gigantica or F. hepatica, and crude proteins were extracted and subjected to SDS-PAGE. Cattle had the preponderance of F. gigantica, while F. hepatica was the dominant species in water buffaloes. ABSTRACT Quail is an important and interesting group of galliform birds. The Common quail (Coturnix coturnix); the Japanese quail (Coturnix japonica); the Panda quail (Coturnix japonica); the Dotted white quail (Coturnix japonica) and the Bobwhite quail (Colinus virginianus) were used in this study. PCR-RFLP and SDS-proteins were performed to reveal the genetic characterization and genetic relationship of the studied quails. Detection of the basement membrane-degrading proteolytic activity of Paracoccidioides brasiliensis after SDS-PAGE using agarose overlays containing Abz-MKALTLQ-EDDnp BJMBR

We have characterized, in the Paracoccidioides brasiliensis yeast phase, an exocellular SH-dependent serine proteinase activity against Abz-MKRLTL-EDDnp and analogous fluorescent-quenched peptides, and showed that it is also active against constituents of the basement membrane in vitro. In the present study, we separated the components of P. brasiliensis culture filtrates by electrophoresis and demonstrated that the serine-thiol exocellular proteinase has a diffuse and heterogeneous migration by SDS-PAGE, localizing in a region between 69 and 43 kDa. Estimation of phylogenetic relationships of Phlomis species based on seed protein polymorphism Electron. J. Biotechnol.

In this study, phylogenetic relationships among 39 Phlomis taxa were investigated based on seed protein profiles produced by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). A total of 21 polypeptide bands were scored, of which, 19 were polymorphic among the taxa of the genus Phlomis. A distance matrix was generated from the similarity matrix which was computed by using Jaccard’s similarity coefficients, based on polymorphic bands and then an UPGMA tree was established through cluster analysis performed on the distance matrix. Growth characteristics and dynamics of protein synthesis in callus cultures from Glycine wightii (Wight & Arn.) Verdc. Ciência e Agrotecnologia

Cotyledon explants were first cultured on MS medium supplemented with 4.52 M 2,4-D and 0.46 mM kinetin. The development of the calli was followed (0, 4, 8, 12, 16, 20, 24, 28 and 32 days after) and the growth curve was determined, based in fresh and dry weight. The growth curve presented sigmoidal form with four distinct phases. The highest growth percentage was observed at the exponential phase and the lowest at the stationary phase. These results indicated that cotyledon callus subculture should be performed 20 days after inoculation. ABSTRACT In this study, we are presenting recommendations to the best agricultural use as well as for plant breeding of three millet cultivars namely ENA1 and ENA2, which have African origin, and BRS1501 originally from India. These cultivars were evaluated for growth, yield and grain quality traits. The morphological traits evaluated in this study indicated that the African genotypes ENA1 and ENA2 are better than the Indian genotype BRS1501 for no-till farming or to produce forage with 15% of crude protein at flowering and at harvest to produce stover (around 7% of crude protein content) for livestock feeding. Cotesia glomerata is a natural enemy of the vegetable plague Ascia monuste orseis and preferably parasites 2nd, 3rd and 4th instar larvae. Parasitism effects on the haemolymph protein profile of Ascia monuste orseis larvae from the 2nd to 7th days were analyzed both qualitatively and quantitatively by SDS-PAGE and Coomassie-Blue binding methods. Quantitative analysis showed a progressive increase in the protein content of about 6.5 and 12.5 times in parasitized and non-parasitized larvae from the 2nd to 5th days, respectively. Improved adsorption-desorption extraction applied to the partial characterization of the antilisterial bacteriocin produced by Carnobacterium maltaromaticum C2 BJM

Bacteriocins are ribosomally produced peptides useful for food biopreservation. An improved adsorption-desorption process is proposed for the partial purification of the bacteriocin produced by the fish isolate Carnobacterium maltaromaticum C2. Analyzis of extract by SDS-PAGE indicated this method may offer an alternative to improve the yield of purification of bacteriocins.

Red scorpions Mesobuthus tamulus (Coconsis, Pocock) were obtained from different regions of West and South India (Ratnagiri, Chiplun and Ahmednagar from Maharashtra and Chennai from Tamil Nadu, respectively). Their venoms composition was analyzed using gel electrophoresis (SDS-PAGE). All venom samples shared six bands of 170, 80, 60, 57, 43, and 38 kDa molecular weights. Bands of 115 kDa and 51.5 kDa were characteristic of venoms obtained from red scorpions of Chiplun region, and the 26kDa band was absent in scorpion venom from Tamil Nadu. Molecular analysis of endophytic bacteria from the genus Bacillus isolated from tropical maize (Zea mays L.) BJM

Endophytic bacteria play an important role in agriculture by improving plant performance and adaptation against biotic and abiotic stresses. In the present study molecular methods were used for identifying Bacillus endophytic bacteria isolated from Brazilian sweet corn. SDS-PAGE of wholecell protein extract of fortytwo isolates revealed a high number of scrutinable bands. Twenty-four isolates were identified in nine different groups of duplicated bacteria and eighteen were identified as unique. Some high-accumulated polipeptides with variable length were observed in almost isolates. The Mycoplasma gallisepticum strains [wild-type S6 (208) and a vaccine-type F-K810] grown in Frey´s and Hayflick´s media were analyzed by SDS-PAGE. No visual changes in the protein profiles of these strains were observed regardless of the media composition used, although the polyacrylamide gel electrophoretograms showed minor differences do exist when densitometer traces of the gel are compared. Both strains were easily differentiated on SDS-PAGE analysis by a peptide band p75, that is specific for MG F-K810 strain, used as vaccine.
